As a South American that worked with Spanish editorials before, I must admit that the differences are not so simple to grasp. Culturally, we think different in the order we communicate our ideas (as every country of course) and the grammar and vocabulary differences are rather plenty as well.
I had a hard time when I used to work as a PR Director even though I was a native speaker, and as of today, I also seem to have difficulty understanding even spanish movies such as: Rec and Orfanato last year.
This gets asked quite a lot. The biggest difference in pronunciation is seseo vs. distinción vs. ceceo, how the C, S and Z are pronounced - Another link. Latin America uses seseo, many places in Spain, particularly the Madrid area, use distinción. That´s not a lisp. They still pronounce the S like and English S, but the soft C and Z like a soft theta.
There´s also a difference in vocabulary, but that´s true between any two given Spanish-speaking areas.
