It depends on your interests. A lot of people start with the video lessons. Some start with posting questions in the How to use this site category or some like to go to the chat room where they can get immediate answers, etc.
I'd suggest hitting them all to start. Especially look through the Reference section and articles like Forum Policies. We have a much overlooked feature called Spanish Media where you can read Spanish newspapers, watch videos, listen to music, etc.
If you can't find it, just ask.
If you're like me (starting with no prior knowledge of the spanish lauguage), lesson 1.1 is a great place to start! Good Luck.
