Saber means to know in the sense of learning something, gaining knowledge, etc. Yo sé que hora es = I know what time it is. Ella sabe hablar portugués = She knows how to speak Portuguese.

Conocer means to know in the sense of being acquainted with, or familiar with. Yo conozco a Juan = I know John. Él conoce Madrid = He's familiar with Madrid.
