Would you use estar or en?

Really: we would use both.

Estoy en la calle (on the street)
Estoy en casa ( at home)

EStoy en el trabajo( at work)

Estoy en la playa (on the beach)

El lapiz está an la mesa (on the table)

La leche está en la taza (in the cup)

As you can see, we get by with just one preposition: EN

So, in case of doubt: location= estar+en

In Spain we use "estar": "Cartagena está en el mar Caribe"
The pronoun "sobre" doesn't sound natural in this context.
"La farmacia está por la calle 44" (we don't know the exact location).
It does in time specifications: "Quedaremos sobre las cinco". We meet at about 5 o'clock.

Se puede decir:
Cartagena queda en en el Mar Caribe
La farmacia queda sobre la calle 44

Pero decir en, en todas las ocasiones, está bien.
