I wouldn't use "el placer es mío" the first time, only if it's the second speaker.

_Juan, ésta es Beatriz.

Juan dice: _ Un placer.
Beatriz responde _ El placer es mío.

I have the feeling that you have to have already used the word "placer" before saying "el placer es mío". "Mío" is emphatic in general and also here.

Another conversation could be: _ Juan, ésta es Beatriz. Juan dice: Un placer or Encantado or Encantado de conocerte etc. Beatriz responde:_ Un placer or Encantada or Encantada de conocerte etc.
