I haven't heard this word in Spanish to refer to a truck, but there is a type of truck which is called a "mule" in English. It is like a golf cart on steroids, with a pick-up bed in the rear. Inasmuch as Mexico is so close to the US, this might just be a new word in the Mexican lexicon.

I should have mentioned this was a conversation taking place between two Colombians. - EKolbinsky, Sep 25, 2016
I found the answer. "Mula" is short for "tractomula," meaning a tractor-trailer. - EKolbinsky, Sep 26, 2016
