Question

There are numerous translations of Of course. Can someone explain them to me?

por supesto
claro
desde luego
de ley

Here's my take on them. Please correct me.

Claro que sí - Of course I'll do what (you/he/she) want me to do! In other words, it has to do with an action.
Por supesto - Agreement. This is used if you agree strongly about what your friend says and believes it to be common sense.
Claro - Dunno...
Desde luego - ?????????

De ley - SpanishDict's 10 Ecuador Slang explains it all:

De ley is used a ton by Ecuadorians, and it means obviously, or of course. A Spanish synonym for it would be claro que sí.

Desde luego. Another of Course.

Also: "¡Desde luego!" (en tono de reproche) For goodness sake!

In some cases it can even mean "Right away!"
