¿Cómo es tu mejor amigo? would ask what your best friend is like- what are their characteristics. Like tall, friendly, studious, etc.
¿Cómo está tu mejor amigo? is a different question that asks how your friend is (at this time)- are they doing well, healthy, etc.
As a fragment in other contexts what you actually wrote might mean something else depending on what is around it, and the punctuation. Please use proper punctuation and capitalization and provide context for the best answers.
