Is there a different meaning or reason there are two different versions?

As far as I know, the correct form would be: agradabilísimo

I have never known of any set of grammar rules governing the structure of superlatives and diminutives - kind of like slang - so you can say it any way you choose, ie: agradabilísimo / agradablísimo / agradablezón / agradablesísimo, etc.

I find it hard to believe that the responsible grammar nerds haven´t come up with a set of rules. Isn´t that what they do? ;) There are a lot published, including there on SD, but I don´t know what is authoritiative, and RAE often just makes my head spin - rogspax, Jul 25, 2013
Again, I have never seen any rules, but how can there be any? In Mexico we say "ito", in Spain "illo", in PR "ico". No matter which you choose, you will always be understood perfectly. - 005faa61, Jul 25, 2013

I rather think that agradablísimo is the correct one. It is one of a fairly small list of irregular superlatives.

I went back and found this, confirming, after I started to doubt myself.

So, I don´t think there are truly, officially, two forms, but rather than the one is an incorrectly (though regularly) formed version of an irregular superlative, akin to goodest.
