¡Que palo! usually means, "What a pain/bummer/downer etc. It might be used to indicate embarrasment as well... as usual that depends on context.
de que palo va
would probably mean something like
"What a downer (bad situation I'm in)
The situation will give you a more accurate translation. What was the situation in which the phrase was used?

One man meets another in order to examine his real intentions and who is he in fact. And he says to third person: "Quiero saber de que palo va." Could it be something like: "I want to know what he is made of"?
