I would have thought it would be as follows.

Es la medianoche y (el número de minutos)

Creo que sería una buena idea decir medianoche porque las doce puede ser medio día..

I asked a native Uruguayan friend this question. He replied that 12:30 am would be given as "las doce y media de la noche." The "de la noche" is added to distinguish the time from early afternoon. They do not use constructions such as "cero y cuarto" or "cero y media."
