When do we use estar and when ser. They both mean to be

Although the use of "ser" and "estar" is a very extensive topic, in basic terms, you could say that "ser" implies permanance or intrinsic values or qualities. "Estar" is used mostly for things of transience, such as moods, feelings, location, most illnesses, etc.

As many would tell you, many times there may be some confusion on which verb to use. For those situations, you will need to supply specific content in your questions.

This is way to complex of a topic to be explained in a couple sentences. In fact people write complete books on it. Your best bet is to educate yourself a little bit on the basics, and then ask specific questions.
