It depends. You usually hear this when you ask someone how they are doing. They will say that they are fine. "¿Y tu?" - and you? So, you can say "Yo tambien" which would mean I am fine too. This is of course assuming that they said they were fine.
If you wanted to say that you were tired instead of saying you were fine, you would respond with "Estoy cansada" for a girl or "Estoy cansado" if you are a boy.
So, there really are many ways that you could answer, but "Yo tambien" is the most common in this most usual instance of the question.

That depends on what the person is asking specifically and what you want to say in reply. "¿Y tu?" as mentioned before means "And you?" Usually this would be said after you asked someone "¿Como esta?" "How are you?" and they replyed something to the effect of "Estoy bien." Once the person replies by asking how you are you could reply "Estoy..." and use any of the following: bien=good, cansada=tired, feliz=happy, triste=sad, mal=not good, etc. All this is in the first lesson of the spanish tutorials. Actually, there is no response to ¨y tu¨ because it means ¨and your¨. ¨Tu¨ without the accent means ¨your¨ It is usually used as a possessive adjective.
On the other hand, ¨tú¨ with the accent means ¨you¨. This is the informal or familiar way of speaking with your friends and your family.
So if my friend and I are asking each other how we are, and she asks me ¨¿Y tú?¨ I would say something like, ¨Estoy bien o estoy cansada o estoy mal.¨
