1.) make = create / produce something. "make me a bicycle, clown!" ... = "Create a bicycle for me, clown!"

2.) do = English auxiliary verb meaning "to perform an action" of some sort.

The meanings are similar but certainly different at the same time. They can't be used interchangeably, but some meanings are similar.

As an auxiliary verb, "do" is used A LOT in English, all the time, in many many different ways.

You can "do" a lot of things, but you can't do people... well, you can, but if you say so you might get some funny looks.

You should clarify, although it can be vulgar, what "doing" a person might mean. I would certainly want to know if I were learning English! - missy8888, Mar 2, 2012
Hi Missy, you're right I should be more clear for English learners, but I didn't want to break any forum rules. Anyhow, lolablues, "to do" someone often implies "to do it," meaning "to have sex." Enjoy! - RedLine55, Mar 2, 2012

Sadly, some of this difference you will have to memorize.

What do you do?
What type of work do you do? Cuál es tu trajabo?

What do you make? How much do you earn at your job?
¿Tu trabajo gana cuanto dinero?

This time of year, my accountant does my taxes.

The baker makes the bread.

I need to do my hair.

I need to make the beds.

This could be the beginning of a game.

Also, in slang (such as in rap songs), people say "I got my hair did" which isn´t proper grammar but means the same as "I got my hair done." - missy8888, Mar 2, 2012

Also, "done" is used to signify when a food is done being baked. "The bread is done" or "The cake is done."
