¿A qué hora vas a descansar? = When (ie:what time) are you going to take a break?
¿Podrías hablar conmigo por un rato durante tu descanso?
= Could you speak to me for a while during your break?
You have not made it clear whether you are asking a boss this question or a friend/colleague...I have made them out as if you were speaking to a friend/colleague because I don't think you would ask the boss in this way..
