¿Será [usted] mi amiga? ¿Será mi amigo?

You can think of the use of Ser as a definition or classification in this context using the CID method.

Your description asks Am I your friend? which is different from your title, but still uses Ser.

Welcome to the SpanishDict forum, Slivia (or should it be Silvia?)

You could also say:

¿Quieres ser mi amigo?

= Do you want to be my friend?

¿Te gustaría ser mi amigo?

Would you like to be my friend?

I think Qfreed has already said in the best way smile

I hope this helpssmile
