What do you want to ask? -- If you want to ask, Where are you from?, then, ¿De donde eres? is correct. If you want to ask, where are you?, then you would have to ask, ¿En donde estás?, ¿En donde está?, ¿Donde estás? or ¿Donde está?

¿De dónde eres? -- Where are you from?
Perhaps you could check out this link:
