I work at a deli counter and have many spanish speaking customers. How would I ask the initial question... "What can I get for You?" or "What would You like?"

And don't forget to say the popular ¿puedo ayudarle en algo? or ¿cómo puedo ayudarlo? Can I help you? How can I help you?

Yes, if you wanted to be polite you could also say

¿En qué puedo ayudarle?

Just to add to the flavor of the answers:

En que le puedo ayudar. Que le puedo ofrecer. Que le gustaría pedir.
