They are synonyms and can often be interchangeable when you want to use the word "again" in English. They might have slightly different connotations, however, in certain circumstances.

Use "otra vez" when you mean "once again" or "one more time".

Use "de nuevo" when you mean "anew" or "all over again".

They're interchangeable. Much like "tal vez" and "quizas" can be used to express "maybe" and "perhaps" or "por supuesto" and "como no" to say "of course."

"de nuevo" means again and "otra vez" means another time. Sometimes they mean the same thing and either one can be used (in English or Spanish) and sometimes they don't.
