Correct. Aside from puede (singular, formal) you can also use puedes (singular, informal), podéis (plural, informal) or pueden (plural, formal) depending on who you're talking to. Also, it's advisable to use the inverted question mark at the beginning of a question in Spanish, thus, "¿Puede ayudarme?"

More informally: ¿Me ayudas? Can you (won't you) help me? Can you help me really means will you help me anyway, and I hear this much more often.

¿Puedes ayudarme?" - can you help me? You could also use: "¿Me puedes ayudar?"
