Un Saludo or Saludos
Through work I receive emails from my Spanish colleagues. Sometimes they sign off "Un Saludo" and sometimes "Saludos". Is there any reason why this is changing - it may be that sometimes an email is sent just to me vs to many people ?

I don't think there is any particular reason for your colleagues using one greeting or the other, it might just depend on their mood. I think it would be equivalent to signing off with either "regards" or "best regards" which is just a minor difference.
