If someone tells ´you are pretty´, you should not answer por supuesto or claro. Unless you want to give the idea you´re a conceited person.
Anyway, as a general rule, I suggest you to think those terms as:
por supuesto --> of course
claro --> sure
-¿Vamos al cine? - ¡Claro!
-Supongo que vendràs a mi cumpleaños. - Por supuesto.
