I think it means, "There is no way.. not even as a joke I'll do it!" But I also knew it as Ni en broma...

Good. Ni de y Ni en broma pueden ser sinónimos (i.e. absolutely not). En broma/de broma by themselves are two different animals. - gone, Jan 28, 2011
