I can safely say that I have probably never heard something like this expressed in the first example; however, if you were trying to construct a sentence using the possessive, you might be able to get away with saying:

My room's air conditioner doesn't work

But even this seems a bit, for lack of a better term, clunky, and I would probably just stick to some variation which included the phrase "in my room."

Both are grammatically correct but the second one (in my room) is used more often.
