Can I just say "Estoy esperándolo" if I'm in the bathroom and someone tries to cut in front of me to use the stall? Gracias.

I think that's be a bit ambiguous. I'd say: "estoy en fila", or "estoy esperando para usarlo".

I concur with Gekkosan: "lo" is fine for people or things that move by themselves (like a bus), but it sounds odd when you mean "waiting for something". While "I am expecting him" is fine when you wait for someone, "I am expecting it" sounds ridiculous when you are expecting to use the bathroom.
