El mejor / la mejor = the best. Lo mejor = the best thing / the best one etc... A lo mejor = maybe, probably etc...
Lo mejor de mí vida eres tú - You are the best thing in my life - I wouldn't say you only use it in a stand alone context.

Note that you only use 'lo mejor' in stand-alone context as per the above.
If you are saying the best something then you use el or la according to the noun.
E.g. Mi amigo Juan Camilo hace la mejor cerveza de Colombia.

"lo mejor" is one of the few neuter gender uses in Spanish.
It can also be used in a completely different context, ie: A lo mejor no va a llover el día de hoy ...... It´s probably not going to rain today.
