Estoy en casa would mean I am a home, that is not used much in Colombian Spanish, rather estoy en mi casa.

Estoy en la casa refers to being in a specific house known to both parties.

Estoy en la clase - here clase pertains to the class room, the people of the class, everything that makes up the class

Estoy en clase refers to being in the process of being taught

No worries. Although there are great differences between when to use definite articles in Spanish and English, in these examples they work the same way. - afowen, Dec 26, 2010

Estoy en casa. - I am at home.

Estoy en clase. - I am in class.

The 'la' is simply not needed in this case... Just as in English, you do not need 'the' here.

Sorry... I only answered one part of the question... afowen gave a good explication of when to use 'la', though. :) - NikkiLR, Dec 26, 2010
