To say "Have a good day" - "Que tenga un buen día", do I need to use "que"? I know that you can use it, just wondering if you must use it. I understand that if I say "I hope you have a good day", I would need to say "que tenga...".

Yes. ¨Que tenga¨ is short for ¨ojalá que tenga¨, so que is setting up the second clause of the sentence. If you want to command them to have a good day, ¨tenga un buen día¨ would come off as more forceful. Adding the ¨que¨ is like saying, ¨may you have a good day¨, or ¨i hope you have a good day¨.
