¿Tienes una novía? if informally.
Or if you want to be formal ¿Tiene una novia?
While "una novia" (no accent) is not wrong, it makes you wonder why are you saying "one" (I know, not in English). How many do you want me to have? When it is not about counting, but about the difference between having and not having, you omit the article:
¿Tienes/tiene novia?
¿Tienes una novia? if informally.
Or if you want to be formal ¿Tiene una novia?
Or you could say "mujer" instead of novia, which means "woman". That might also be construed as "wife", but if you are trying to find out for yourself, that might not be a bad choice.
Context is very import here because aside from "Girlfriend," "Novia" also means "Fiancée."
