Neither of them. What about an accent?
Your first translation could mean "Your father is an illness"
Your second "Your father that illness"

I am not sure but I think that ( estar ) will be better because when you describe your self you use ( ser ) but those descriptions are Permanent , but I don't think that being sick is Permanent
