"Vamos" is from the verb ir. "Vámonos" is from the verb irse.
"Vámonos" is a command conjugated for nosotros and experiences an odd spelling change. It's a combo of "vamos" + "nos" just like "Vete" is a combo of "ve" + "te." When you have nosotros commands that are also reflexive (like with irse, dormirse, etc.), the s is taken off. Just as with any command that attaches pronouns, an accent is used to express where the accent is -- like with many words in general.
Using that, think of what the difference between ir and irse is, and you should have a good idea of how to use them.
Both phrases are used interchangeably when you want to say "Let's go."
However, you would use "Vamos" in a sentence such as "Vamos al cine" (We are going to the theater.)
