How would I say something such as, " She is the prettiest of them all."

And then also something like, "I am the youngest in the family".

You do not use más with mejor and peor, right?

Simplest of all - "lo mas sencillo de todo". "Mas" is "most" so just slip it in before your adjective.

Prettiest = mas bonita

Youngest = mas joven

lots of etc.

... and as Gekkosan pointed out the other day you no longer even need to put an accent on "Más". Wow!
