How would I say it is 4:40? Do I say son las cinco menos veinte or son las cuatro y cuarenta?

Se puede decir los dos! You can say both, they both work just fine. I would say that "son las cinco menos veinte" would be more common.

Son las cinco menos veinte since it's in the second half of the hour but you'd be understood either way.
