I am en-route
Could somebody explain how I would say "I am on my way" as in "en-route" please. Dictionary does not give equivalent.

Act I - Place you cursor on the word route in your question above. Double-Click on the word route. In the narrow blue window that opens, single click on the word route.
Act II - Place you cursor on the word ruta in Item 1. of the definition of "route". Double-Click on the word ruta. In the narrow blue window that opens, single click on the word ruta.
All that just to see that:
en ruta (hacia) -> en route (to)
But the more interesting thing is your question.
This is how you asked your question -> how would I (Caza) say "I am on my way" as in "en-route". If you were "en-route" you would not be there to answer the telephone unless you were called on a portable (cell) phone. So, you really need 2 answers at the ready. One answer if you are in fact on your way out the door and answering a fixed in-house telephone, and, another answer if you are travelling and answering a cell phone. So think about these replies.
1. You are still in the villa east of Malaga (I am envious) and answering a home phone:
Quick and easy -> Ya voy = I'm coming
A little longer -> Me yendo ahora mismo = I'm leaving right now., o
Also longer, --> Me voy ahora mismo = I'm leaving right now
2. You are underway and answering a cell phone:
Estoy en ruta ahora mismo = I'm en route right now
