When answering questions in a negative command. how do you determine the ending of the word? if you put lo or la or plural endings?

With negative commands the direct object pronoun (lo or la) would precede the verb so you should not be confused any more than when conjugating any other verb.

No me lo das. (negative, familiar commands use 2nd person, singular, present subjunctive endings)

No me lo dé. (negative, formal commands use 3rd person, singular, present tense, subjunctive endings)

Don't give it to me.

I don't understand the part about "when answering questions."
