What is the difference between

1: "Que talla usa usted?"

and

2: "Que numero usa usted?"

Is talla clothing size and numero shoe size?

...So would the answer for #1 be "Uso la diez." and for #2 be "Uso el ocho." ?

You are correct. "Talla" is used for clothing size, and "número" for shoe size.
